Strain developmentProduced by corssing relA deficient mice and TNF deficient mice strains. Neomycin cassettes were inserted into the nucleotides 3704-5364 of TNF gene and the downstream of the translation initiation codon of relA gene, respectively. RelA deficient mice were backcrossed to C57BL/6 for 10 generations, and TNF deficient mice were backcrossed to C57BL/6 for 5 generations. Then, the two mutant strains were crossed to create the TNFkB mice. Developed by Takahiro Doi at Aichi Cancer Research Center in 1997.
Strain descriptionTNF and RelA double knockout mice. TNF, tumor necrosis factor is a multifunction proinflammatory cytokine. RelA, v-rel reticuloendotheliosis viral oncogene homolog A (Avian) is one of NF-kB family relating to the development of lymphocytes, suggested to be highly sensitive to TNF cytotoxicity. Mice lacking only relA gene die between days 14 and 15 of embryogenesis due to liver cell apoptosis by endogenous TNF cytotoxicity. Meanwhile, TNF/relA double-deficient mice are viable and have normal liver. This strain is useful for studies of functions of RelA in vivo.
