Strain developmentTargeting of Stealth: 2001, Denis-Luc Ardiet, ISREC
Strain descriptionMice with a lacZ knock-in in the first major coding exon of Stealth. The lacZ ORF begins with the first start codon of that stealth exon. Purpose of that mouse strain is to analyze stealth function in mice, in particular for its role in development, innate immunity, and homeostasis of organ systems.PGKneoR cassette flanked with loxP sites still present in this stealth null allele because these mice haven't been crossed with a deleter strain since there hasn't been one isogenic for 129SvEv(Allan Bradley).
Colony maintenanceThis strain has been kept isogenic for the genetic background 129SvEv (source: Allan Bradley) that is the very same genetic background of the ES cells in which the Stealth null allele was generated by a knock-in of lacZ in frame wih the major first start codon of Stealth. Hence, this precious isogenic line shall be kept breeding in its literally original background. In particular, the Stealth null allele is homozygous viable when isogenic for 129SvEv. However, such homozygotes don't breed, become sick with age and develop so far uncharacterized inflammatory processes and cachexia.
