Strain development The expression construct was created by cloning cDNAs encoding the selected genes and the promoter into a plasmid vector. After verification of the construct by nucleotide sequencing, the transcriptional unit was cleaved out with restriction enzymes, purified with biochemical method and used for microinjection. The offspring were initially examined by PCR genotyping and in vitro expression of the marker, EGFP, after transfection with Tet-OFF expression vector (Clontech). The entire process took from 1999 to 2003.  
Strain description The transcriptional unit integrated into PTR-H consists of a dominant negative form of TGF-beta type II receptor gene and EGFP marker under a bidirectional tetracycline-responsive promoter. By crossing with another transgenic mouse that express tetracycline-inducible transcriptional activator, TGF-beta transduction pathway can be disrupted in a conditional manner.
